چکیده
An indigenous locally isolated white rot fungal strain Phanerochaete chrysosporium IBL-03 was investigated for the hyper-production of ligninolytic enzymes from different agro-industrial wastes including wheat straw, rice straw, banana stalks, corncobs, corn stover, and sugarcane bagasse as substrate material in still culture fermentation technique. Screening experiments were performed at 30C from 1 to 10 days and maximum enzyme activities were recorded after the 5 day of incubation on banana stalk. P. chrysosporium IBL-03 produced highest activities of lignin peroxidase (LiP) and manganese peroxidase (MnP) but no laccase activity was detected in any fermented culture media. Production of ligninolytic enzymes was substantially enhanced through the optimization process. When banana stalk at 66.6 % moisture level and pH 4.5 was inoculated with 5mL spore suspension of P. chrysosporium IBL-03 at 35C in the presence of molasses (1%) as carbon source, ammonium sulfate (0.2%) as nitrogen supplement, (1%) Tween-80 (0.3 mL) as surfactant and mediators (MnSO4 and veratryl alcohol) enhanced the LiP and MnP production up to 1040 and 965 (U/mL), respectively.

-Among 12 basidiomycetaceous fungal strains, Phanerochaete chrysosporium SC26 and K3 and Phanerochaete flavido-alba FPL 106507 were the most efficient in decolorizing paper mill wastes. Enzymatic activities attributable to both families of extracellular ligninases, lignin peroxidase (LIP) and manganese peroxidase (MnP), have been detected in P. flavido-alba FPL 106507. Both peroxidase activitie...
متن کاملExpression analysis of extracellular proteins from Phanerochaete chrysosporium grown on different liquid and solid substrates
White-rot fungi secret a large number of hydrolytic and oxidative enzymes for degradation of lignocellulosic material. The sequencing of the genome of the white-rot fungus Phanerochaete chrysosporium has facilitated the characterization of its complete extracellular proteome. P. chrysosporium was grown on liquid medium, containing glucose, cellulose or wood chips as the carbon source, and also ...
متن کاملEnvironmental Factors and Bioremediation of Xenobiotics Using White Rot Fungi
This review provides background information on the importance of bioremediation approaches. It describes the roles of fungi, specifically white rot fungi, and their extracellular enzymes, laccases, ligninases, and peroxidises, in the degradation of xenobiotic compounds such as single and mixtures of pesticides. We discuss the importance of abiotic factors such as water potential, temperature, a...

One of the most economically viable processes for the bioconversion of many lignocellulosic waste is represented by white rot fungi. Phanerochaete chrysosporium is one of the important commercially cultivated fungi which exhibit varying abilities to utilize different lignocellulosic as growth substrate. Examination of the lignocellulolytic enzyme profiles of the two organisms Phanerochaete chry...

BACKGROUND The genome of Schizophyllum commune encodes a diverse repertoire of degradative enzymes for plant cell wall breakdown. Recent comparative genomics study suggests that this wood decayer likely has a mode of biodegradation distinct from the well-established white-rot/brown-rot models. However, much about the extracellular enzyme system secreted by S. commune during lignocellulose decon...
